Abstract:
An apple bacterial artificial chromosome (BAC) library, consisting of 35,328 clones with an average insert size of approximately 110 kb, was constructed from the apple cultivar ‘GoldRush’. By assuming that the apple genome is 769Mb per haploid in size, the library represents about five times apple haploid genome equivalents.
This suggests that there is a 99% probability of finding any specific apple sequence in this library.
PCR-based screening of the library was carried out using four random sequence characterized amplified regions (SCARs) as well as 11 Vf-linked SCAR markers.
On average, 6 positive BAC clones per random SCAR and 2.6 per Vf-linked SCAR marker were detected, respectively.
Therefore, the library is desirable for map-based cloning of the Vf gene, and for other genes controlling horticulturay important traits.
In addition, it is useful for constructing a physical map of the apple genome.
