Abstract:
A method for the in vitro initiation, multiplication and rooting of explants from L. ‘Sunrise’ is described.
Pretreatments of mother plants with 200 mg/l benzyladenine considerably increased the number of buds per branches. However, no significant differences on growth and multiplication between pretreated and non-pretreated explants were observed during in vitro establishment and multiplication.
Nodal explants segments (1.5-2.0 cm in length) were excised from the branches and induced to form axillary buds on a modified Murashige and Skoog Medium) at half strength (½ MS) and supplemented with 1 mg/l benzyladenine, 3% sucrose and 0.7% agar.
The response of explants during the establishment phase depended on collection period. The best results were obtained during spring.
The medium used for multiplication phase was ½ MS with 0.2 mg/l BA, 3% sucrose and 0.7% agar. A multiplication rate about 4.0 was achieved.
The use of a double-phase system did not improve the multiplication rate. Microshoots formed in vitro could be rooted under aseptic conditions.
The major problem encountered in this study was a low rooting percentage.
About 35.3% of the microshoots could be rooted by culture on the same basal medium supplemented with 2 mg/l indolebutyric acid for three days and further subculture in a growth regulator-free medium.
