ISOZYME CHARACTERIZATION OF OPUNTIA SPECIES AND VARIETIES FROM ITALIAN GERMPLASM

Abstract:
In Opuntia only a few studies have been conducted on genotypes description, mainly based on morphological data. The purpose of this paper is the genotypes identification by electrophoretic analysis of Opuntia species, varieties commercially grown for fruit production in Italy and biotypes selected within the Sardinian and Sicilian germplasms. Among the different tissues of the plant tested (root, cladode, petal and pollen) the best results have been obtained with pollen, that evidenced activity of the thirteen enzymes examined. The isozyme patterns of nine genotypes of the Sardinian germplasm, fourteen of the Sicilian germplasm, ten of Sicilian origin but grown in Sardinia and eleven genotypes of species different from Opuntia ficus-indica have been analysed. MDH enzyme was the most effective for identifying genetic variability within the population tested, both on cladodes and pollen.
