The present study was designed to elaborate a single step method for analyzing phenolics in plant extracts containing ellagitannins of different molecular weights.
Extracts of strawberry, raspberry, blackberry and related Rosaceous plants were analyzed by HPLC on silica gel RP-18 with three-wavelength UV/VIS detection.
With common gradient HPLC methods, the ellagitannins sanguiin H-6 and lambertianin C of raspberry and blackberry leaves were not resolved.
On the other hand, the complete phenolic pattern could not be elaborated by a set of isocratic HPLC runs, because of changing peak sequence due to elutional solvent strength and variable S-values of the phenolic components.
Conditions for a successful gradient separation of sanguiin H-6 and lambertianin C were: Minimum isocratic dwell time of 7 minutes and shallow steepness of the gradient to specifically retard the larger molecule lambertianin C. The present method could be applicable in studies investigating the relationship between phenol content, physiological ageing of plant organs and resistance to pathogenic fungi and plant viruses.
