Using the analytical hierarchy process to select the most appropriate method for grapevine Pinot gris virus and grapevine fleck virus elimination

Abstract:
This study aimed to identify the most effective methods and parameters for eliminating co-infections of grapevine Pinot gris virus (GPGV) and grapevine fleck virus (GFkV) in grapevine. We assessed various techniques including meristem culture, intensely regenerative apex culture, in vitro thermotherapy, in vitro chemotherapy, and electrotherapy (through electrical stimulation in a uniform electric field and in a horizontal electrophoresis tank). The Analytic Hierarchy Process (AHP) was employed to rank these methods based on their virus removal efficacy and economic efficiency. The biological material for this study comprised V. vinifera L. genotypes ‘Sangiovese’ and ‘Negru mare’, diagnosed with GPGV and GFkV viral complex. Addressing mixed viral infections is crucial, as each virus in the complex impacts the health of the regenerated plants, which must be free from both pathogens. The AHP analysis, conducted using SpiceLogic Analytic Hierarchy Process software, indicated that in vitro chemotherapy was the most effective method for eliminating GPGV, significantly outperforming meristem culture, apex culture, electrotherapy and, in vitro thermotherapy.
