Effects of temperatures on seed germination and initial growth of seedlings in Wisteria floribunda

Abstract:
Wisteria floribunda is a liana woody plant distributed in Japan. This plant bears long beautiful racemes in spring, and the flowers grow into seedpods in autumn. Although vegetative propagation is usually employed in commercial production, there is less information of sexual reproduction in this plant. Thus, this study was conducted to clarify the optimum incubation temperatures for seed germination and initial growth of seedlings in W. floribunda, and methods of breaking dormancy of seedlings was also examined. Seedpods were collected from 9 plants of W. floribunda. After water absorption and surface sterilization, the seeds were incubated at 15, 20, 25°C under dark, and number of germinated seeds was recorded every four days. The seedlings were cultivated at the same temperatures as seed germination under natural light. At 100 days after transplanting, a half of the seedlings in each treatment continued to be grown at same the temperature, and other half were grown at 15°C for the first month, followed by one month each at 4 and 15°C, and then returned to the original temperature. Number of seedlings forming new branch was counted after each treatment. Initiation of seed germination at 25°C was earlier than 15 and 20°C, whereas the difference of seed germination among temperature was not finally recognized. New branch and leaf development were not observed at 100 days after transplanting. Seedlings that were exposed to cold treatment accelerate new shoot and leaf developments at 20 and 25°C, and both seedlings treated with cold temperature and non-treated showed these developments at 15°C. From these results, seed germination was similar among different temperatures from 15 to 25°C. In addition, it was estimated that cold treatment is effective to break bud dormancy of seedling.
