Advance of adventitious root formation and production, and saponin accumulation by in vitro cultures of Giao Co Lam (Gynostemma pentaphyllum)

Abstract:
Giao Co Lam (Gynostemma pentaphyllum) is a rare valuable medicinal herb used for the human being's health as it has such medical functions as anti-stress properties, restoring the body's balance, improving memory, preventing thrombosis, inhibiting growth of tumors, and treating diabetes, etc. However, that productivity is still limited and could not meet consumer requirements. Nowadays, the development of plant cell technology has opened up a new way to solve some problems in the field of cultivation. In particular, the adventitious root induction from in vitro culture for biomass collection is considered as an effective technique. The in vitro leaf explants from 42-day-old in vitro plantlets cultured on Murashige and Skoog (MS) medium supplemented with napthalene acetic acid (NAA) at 1.5 mg L-1 induced callus with the highest rate (77.78%). The rate of root-induced calli was optimal on MS medium added with indolebutyric acid (IBA) 0.5 mg L-1 and reached 91.11%. Schenk and Hildebrandt (SH) medium added with IBA 0.5 mg L-1 and NAA 0.5 mg L-1 was suitable for proliferation of root biomass in liquid medium with the highest weight of root biomass at day 28 of the culture. The supplementation of methyl jasmonate (MJ) into the cultures did not increase the proliferation of root biomass but it enhanced saponin content. MJ at 10 mg L-1 increased the saponin content of root biomass at the highest level.
