Effects of CLE peptides on growth of in vitro roots and shoots of persimmon

Abstract:
Peptides are short amino acid chains, and peptide hormones such as growth hormone, insulin, and vasopressin are well known as important hormones in animals. Peptides also exist in plants, and in recent years many studies have demonstrated that peptide signalling plays a great role in various aspects of plant growth and development. Arabidopsis thaliana has 32 CLE peptides, named for the CLAVATA3 (CLV3)/ESR-related peptide family, including CLV3. The CLE peptides are thought to regulate cellular differentiation activity in the shoot and root apical meristem, and synthetic CLV3 peptide reduced the size of shoot and root meristems. In this work, CLE25, a member of the CLE peptide family, was added to the root-inducing medium at 1 ÁM and found to inhibit growth of 'Hiratanenashi' persimmon root. The treatments after the formation of root primordia were more effective than those at the start of the root induction. The roots treated with CLE25 10 days after the root induction were significantly thinner and shorter than the non-treated roots. Microscopic observations revealed that the cell division of the cortical tissue in the treated roots was inhibited. CLV3 was also found to have the same effects as those of CLE25. However, all the treatments of CLE25 to the shoot apical meristems, that is, immersing the shoot tips in the agar medium, putting the agarose medium on the shoot apexes, and culturing in the double-phase medium were not effective.
