DNA fingerprinting in rice (Oryza sativa L.) with transposon markers

Abstract:
Rice is one of the most important model plant widely used for genetics studies. Approximately 35% of rice genome consists of mobile genetic elements. Our aim was to analyse Houba (Tos5/Osr13), RIRE1, Hopi (Osr27) and Osr30 retrotransposons movements in rice with IRAP marker technique. These retrotransposons were selected because they are the most abundant retrotransposons in rice. Polymorphism ratios were calculated with Jaccard similarity index. Results showed that rice samples had a variable band profiles. Polymorphism ratios were 75% for Hopi, 57% for Osr30, 52% for Houba, and 45% for RIRE1. In addition to polymorphisms, we also obtained homomorphic bands. These results could be expected to identify rice varieties by using retrotransposon-based markers.
