FRACTIONATION OF ACTIVE COMPONENTS FROM PIPER CF. FRAGILE ESSENTIAL OIL AS AROMATHERAPY FOR ANTI-OBESITY

Abstract:
The purposes of this research were to find constituents of Piper cf. fragile essential oil and evaluate their active components as aromatherapy for anti-obesity using in vivo experiment. Four fractions were obtained from fractionation of ‘sirih merah’ crude oil in silica gel column chromatography. Fractions 1 and 2 had more intense aroma than Fraction 3 and 4. Fraction 1 contained monoterpene and sesquiterpene compounds, while major component in Fraction 2 was monoterpene alcohol compound. The antiobesity activity of each crude oil, Fractions 1 and 2 were evaluated in vivo in male Sprague dawley mice by aromatherapy and inhalation technique. Forty male Sprague dawley mice were used and divided into five groups namely; normal group, consuming normal diet; negative group consuming high cholesterol diet without treatment; crude oil, Fraction 1 and Fraction 2 groups were treated with high cholesterol diet followed by inhalation of crude essential oil, Fraction 1 and Fraction 2, respectively. The result showed that monoterpene and sesquiterpene compounds in crude oil and Fraction 1 were able to decrease body weight of mice. On the other hand, alcohol monoterpene compound in Fraction 2 showed opposite effect, it increased body weight of Sprague dawley mice above the negative group. In addition, the crude oil group showed the lowest cholesterol and triglyceride level in blood compared to other groups significantly (P<0.05).
