THE MICROPROPAGATION AND IN VITRO MAINTENANCE OF SOME PLUM CULTIVARS

Abstract:
In vitro propagation of plum cultivars ‘Anna Späth’, ‘Carpatin’, ‘Dany’, ‘Ivan’, ’Jubileu 50’, ‘Althanns Reneklode’ and ‘Stanley’ was carried out in order to test the in vitro conservation and screenhouse maintenance of virus-free material. The initiation of the in vitro cultures was done in September 2009 from shoots belonging to virus free plants from the RSFG Bistrita. Murashige & Skoog 1962 (MS) + 0.7 mg/L 6-benzylaminopurine (BAP) medium was used both for culture initiation and for multiplication. As growth regulators, various concentrations of BAP, zeatin and gibberellic acid (GA3) were used. For rooting, MS basal medium was used and additional 1 mg/L 3-indolylbutyric acid (IBA) and ex vitro acclimation was done in hydroculture.
