IN VITRO MORPHOGENIC POTENTIAL OF TWO GENOTYPES OF COWPEA (VIGNA UNGUILATA L. WALP.)

Abstract:
Cowpea (Vigna unguiculata L. Walp.) is an important legume food crop that is commonly grown in Paraíba and other states from the Brazilian semi-arid region. In addition it has a strong social and economic importance although it has low productivity. The application of biotechnological approaches for the improvement of cowpea genotypes is currently not possible due to the lack of a regeneration and transformation system. Therefore, comparatively little work has been done to develop and optimize regeneration and transformation procedures, including recalcitrant cereals. The goal of our research was to compare in vitro morphogenic potential of two genotypes, ‘Rabo-de-tatu’ and ‘Branco’ from cowpea [Vigna unguiculata (L.) Walp.]. Experiments were performed in 2×5 factorial completely randomized design combining two cultivars and five concentrations of NAA or BA, using as explant source hypocotyl segments, shoot tips and apical plumule, cultured in Murashige and Skoog (1962) medium. Data were subjected to analysis of variance by F test at 5% probability. The best regenerative performance was ‘Rabo-de-tatu’ genotype. Shoot tips was the best explant for both genotypes. Addition of NAA and BA (above 0.25 mg L-1) inhibited the frequency of shoot regeneration when apical plumule was used as a source of explant.
