SOME INFLUENCES OF α-NAPHTYLACETIC ACID ON THE DIFFERENTIATION OF MERISTEMS OF LILIUM SPECIOSUM 'RUBRUM' NR. 10 IN VITRO

Abstract:
Meristems adventitiously formed on 0.5 cm2 pieces of bulb-scales of Lilium speciosum 'rubrum' nr. 10 were cut from the scale tissue and cultured on nutrient media consisting of MS salts, LS organics, 3% sucrose, 0.6% agar and various concentrations of the auxin NAA (0.00 – 1.00 mg/l). After 10 weeks of culture under controlled light (TL 33; 5.2 W/m2) and temperature (21° ± 1°C) conditions, differences were found in the number of roots, outgrowth of leaf initials, bulblet growth and sprouting behaviour. Higher NAA concentrations stimulated rooting and sprouting and inhibited outgrowth of the primordia. An optimum in bulb growth was found between 0.03 – 0.10 mg NAA/l. The differences in sprouting behaviour of plantlets grown on the various media persisted after planting in soil, but were influenced by a pre-planting cold treatment.

The role of NAA in the various processes is discussed.
