VIROIDS ON PETUNIA AND OTHER SOLANACEOUS CROPS IN THE CZECH REPUBLIC

Abstract:
During the year 2007 the Czech Plant Protection Service carried out an extensive survey for Potato spindle tuber viroid (PSTVd) on ornamental solanaceous crops. The survey was targeted at Solanum jasminoides and Brugmansia species primarily. At the locations of positive findings, subsequent inspection of other possible solanaceous hosts was carried out. RNA was extracted from the leaf samples and the approximately 360 bp long products were amplified by RT-PCR using PSTVd primers and sequenced. A total of 97 samples from 36 locations were tested, including 52 samples of S. jasminoides and 45 samples of other species (e.g. Brugmansia spp., Solanum rantonnetii, Petunia spp., Lycopersicon esculentum, Solanum muricatum). PSTVd was detected at 25 locations, mainly on S. jasminoides host plants. The other 3 species positively tested for PSTVd were Petunia × hybrid (vegetatively propagated) samples collected from two growers, one sample of Brugmansia and one sample of Solanum muricatum. This is the first finding of PSTVd on petunias and the first finding of PSTVd on Brugmansia spp. and S. muricatum in the Czech Republic. Petunia samples, collected from three sites, gave an RT-PCR product of approximately the expected size for PSTVd (± 360 bp), but the sequenced amplicons were shown to be isolates of Tomato chlorotic dwarf viroid (TCDVd). This is also the first finding of TCDVd in the Czech Republic.
