MODULATION OF LIPID TRANSFER PROTEINS ALTERS CLUBROOT DEVELOPMENT IN ARABIDOPSIS THALIANA

Abstract:
The clubroot disease is caused by the obligate biotrophic pathogen Plasmodiophora brassicae. It is world-wide one of the most damaging diseases within the familiy of Brassicaceae which includes many commercially important agricultural crops like cabbage, radish and oilseed rape. To analyse changes in the plant during the interaction, we have performed expression analyses using the ATH1 Affymetrix 22K microarray and RT-PCR. The results demonstrated that plant lipid transfer protein (LTP) genes were differentially expressed during clubroot development. To study the function of lipid transfer proteins during disease development we have generated transgenic Arabidopsis thaliana plants that counteract the naturally occurring regulation in infected roots using a root specific promoter. LTP-Knock out lines were also analysed. So far, we have shown that knock out or over-expression of one LTP gene modulated the expression of other LTP genes. LTP over-expression resulted in decreased susceptibility to clubroot whereas decreased LTP expression led to a higher susceptibility.
