ISHS Acta Horticulturae 812: III International Symposium on Acclimatization and Establishment of Micropropagated Plants

EFFECTS OF CHILLING AND HORMONAL SUPPLY ON ROOTING AND IN VIVO ESTABLISHMENT OF MICROPROPAGATED PLANTLETS OF HELLEBORUS SPP.

Authors:   M. Beruto, P. Curir
Keywords:   acclimatization, auxin, ex vitro, kinetin, micropropagation, rooting
Abstract:
In this study, we have studied rooting and acclimatization of micropropagated Helleborus shoots. H. niger and a number of H. × nigercors genotypes were in vitro multiplied for a period of 12 months. The conditions during the final propagation cycle had a marked effect on the rootability. Multiplication on MS-based medium and with increasing kinetin concentrations enhanced rootability. Shoots pre-treated at 7°C in a solution containing IBA and NAA for 7 days and subsequently cultured on gelled media with no hormones gave satisfactory rooting and in vivo survival (87 and 85%, respectively).
