Abstract:
Using stem explants excised from seedlings germinated in vitro, efficient plants regeneration via organogenesis was established for Trichosanthes kirilowii. To establish optimal conditions for adventitious shoot buds induction, varieties of explants were prepared from seedlings and there were cultured using several media containing different concentration of cytokinins.
Multiple shoot buds, without an intervening callus phase, were induced from stem explants on Murashige and Skoog (MS) medium containing 2 mg L-1 N6-benzylaminopurine (BA). The culture medium, carbon source as well as plant growth regulators were found to influence further shoot multiplication and eventual shoot growth.
Adventitious shoots were subcultured on elongation medium composed of MS medium with 0.5 mg L-1 BA and the elongated shoots were successfully rooted on half-strength MS medium without plant growth regulators.
Regenerated plants showed 98% survival after acclimatization in soil and were morphologically similar to the source plant under field conditions.
