INFLUENCE OF OSMOTIC STRESS ON BIOCHEMICAL PROPERTIES IN POTATO

Abstract:
Drought is among the most limiting factors for plant productivity. Potato is a drought-susceptible crop, however there are differences in the susceptibiliy to water stress between cultivars. It is important to understand how plants respond and adapt to such kinds of stresses. The problem is that there is not enough information available about biochemical markers. Such markers are supposedly present, and they probably change in quantity as a function of the stress, but they have not yet been studied in vitro. Three potato clones with different drought tolerance were examined as in vitro plantlets (‘Boró’, ‘Réka’, 77365/103) and in callus-tests (‘Boró’, ‘Réka’, 736/82). Plantlets were grown on MS medium supplemented with mannitol (0.2 M and 0.4 M), and calluses were grown on 0.8 M mannitol added to MS medium. Among the changes we tested the activity of the enzymes peroxidase (POD) and polyphenol oxidase (PPO) and the total phenol content. The results confirm that the tested parameters are influenced by both the genotype and the degree of the stress.
