Abstract:
Gerbera is one of the best cut flowers in the world.
In this research, effect of thidiazuron (TDZ) on shoot production from young capitulum was studied for first time.
The explants were cut from young capitulum of two Gerbera cultivars (‘Jaimy’ and ‘Mammut’), disinfected and cultured on basal MS (Murashige and Skoog, 1962) medium with different concentration of TDZ (0.01, 0.1, 0.5 and 1 mg L-1). Experiment tubes were incubated in darkness at temperature of 21±2°C for one month.
Then the experiment tubes were incubated under irradiance of 40 μmol m-2 s-1 with 16 h photoperiod and temperature of 25±2°C. The highest number of shoots was obtained on medium supplemented with 0.5 and 0.1 mg L-1 TDZ with 4.5 shoots/explant.
The highest number of shoot was obtained from ‘Jaimy’ than ‘Mammut’ cultivar.
The highest shoots were observed on 1 mg L-1 TDZ (mean height 2.6 cm) and the lowest on medium supplemented with 0.01 mg L-1 TDZ (0.85 cm).
