ISHS Acta Horticulturae 812: III International Symposium on Acclimatization and Establishment of Micropropagated Plants

INFLUENCE OF EXPLANT TYPE, SUCROSE AND IBA ON IN VITRO GROWTH OF ACER PLATANOIDES L. 'JIRKA'

Author:   J. Šedivá
Keywords:   micropropagation, rooting
Abstract:
Experiments were conducted to examine the effect of sucrose levels and explant type on regeneration of Acer platanoides ‘Jirka’ and the effect of indole-3-butyric acid (IBA) levels on root formation. In vitro cultures were derived from mature trees (5-year old). Reduction of sucrose concentration from 30 to 20 g.L-1 caused elongation of shoots and lower callus induction. Presence of IBA in medium increased root induction and number of roots. The maximum of 63% rooting was obtained on half WPM medium supplemented with 10 mg.L-1 IBA. Plantlets were successfully transferred to pots and acclimatized.
