Abstract:
In vitro propagation protocol of Stemona tuberosa Lour., an antitussive medicinal herb was developed using shoot culture.
Shoots were induced from in vitro shoot and node explants in solid MS (Murashige and Skoog, 1962) medium supplemented with 20 μM BAP. Two types of media, B5 and MS, and different concentrations of MS media were tested for the screening of suitable media.
Amongst them, full strength of MS was the most effective for in vitro culture.
For the step of shoot multiplication, shoots were cultured in MS media supplemented with various types and concentrations of cytokinins.
Sixteen combinations of BAP (0-20 μM) and IBA (0-5 μM), were also tested.
Shoot numbers from explants were examined in all media.
The highest number of shoot multiplication rate could be observed on solid MS medium containing 20 μM BAP. Shoots were cultured on MS media supplemented with different types and concentration of auxins for root induction.
Regenerated shoots were successfully rooted after cultured on solid MS medium which were without plant growth regulators for 2 months.
Plantlets could be established and grown well in the greenhouse.
