INFLUENCES OF MEDIA AND CYTOKININS ON SHOOT PROLIFERATION OF 'BRIGHTWELL' AND 'CHOICE' BLUEBERRIES IN VITRO

Abstract:
Nodal segments from mature field grown plants of ‘Brightwell’ and ‘Choice’ rabbiteye blueberries (Vaccinium ashei Reade; syn V. virgatum Ait.) were established on WPM with 4 mg•l-1zeatin. Shoot growth was evaluated on six different media including Anderson medium, WPM, a modified WPM (MWPM), MS medium, a modified MS medium (MMS) and Lyrene’s medium. For 'Brightwell', explants Anderson medium, MMS medium and WPM produced the most shoots after ten weeks culture, but the status of shoots was better on MMS medium and WPM. The highest multiplication rates of total shoots and usable shoots were observed of 'Choice' on MWPM, which reached 8.42 total and 5.83 useable shoots / explant. The effect of three cytokinins at a range of concentrations on shoot multiplication rate was compared. Zeatin at 2 mg•l-1 was most effective in promoting shoot proliferation in both cultivars. Microshoots of 'Brightwell' were induced to root in a perlite-peat mixture after dipping indole-3-butyric solution under intermittent mist and the rooting percentage reached 91.6%. Rooted plants were successfully moved to soil and grown in pots.
