Abstract:
In order to analyze relations in the genus Pyrus, nucleotide sequences from non-coding regions of TrnL-TrnF region of chloroplast DNAs (cp DNAs) were obtained for 44 pear samples including 31 Iranian pear species and cultivars.
Other cultivar samples including 'Yali', 'Housui', 'Chojuro', 'La France', 'Bartlett', 'Balixiang' and 'Okusankich' were also used.
African species P. mamorensis and P. gharbiana, two European species, P. amygdaliformis and P. pyraster and the cultivar 'Golden Delicious' apple were integrated in the current study.
Sequencing and alignment of chloroplast DNA indicated single nucleotide deletion and insertion in DNA sequence of amplified region.
Deletion of TAATTGAC at position 367-374 bp was observed for few samples including acessions of Natanzi, Jose Ghand, Housui, Chojuro, Okusankich and Yali.
The changes of G or A to T were observed in different site length of TrnL-TrnF region for different Iranian pear samples.
Single nucleotide insertion of T was observed for 5 Iranian pear samples in position of 403 bp.
One change of G to T in 150 and A to T in 401 was observed.
One cladogram was constructed from Wagner parsimony analysis.
The most parsimonious cladogram consisted of four major clades.
The first clade included Iranian pear cultivars Natanzi and Jose Ghand and the Japanese cultivar Housui and the Chinese cultivar Yali.
The second clade consisted of Shah Miveh Mashahad, Laleh cultivars and some specimen of P. syriaca species.
The remaining specimens constructed the third clade which harbored different genotypes of P. communis. The fourth clade consisted of P. amygdaliformis and two African species P. mamorensis and P. gharbiana.
In conclusion, some Iranian cultivars presented close relationship with Housui and Yali cultivars in one clade and with some other P. communis genotypes in another clade.
