Abstract:
In vitro regeneration of olive cv.
Rowghani has been studied.
Micropropagation was initiated by using explants from embryos and seedlings.
Sterilized embryos were germinated in OM medium supplemented with 2iP (0.5 mg/L). After 2 weeks, plantlets were sub cultured in OM or Leva media with 2iP (0.5 mg/L). In later step OM medium with BAP (1 mg/L) and 2iP (1 mg/L) were used for axillary bud stimulation and subsequent shoot multiplication.
The carbohydrate sources were sucrose (30 g/L) or mannitol (18 g/L). The results showed both sucrose and mannitol were equally suitable for plantlet growth in the first subculture.
Although, the results of axillary buds cultured in OM medium with sucrose in comparison with mannitol, increased shoot proliferation and improved the general quality and uniformity of shoot cultures, but the number of shoot branches was increased by mannitol.
