Abstract:
Anthers of Dianthus chinensis L. and D. barbatus L. containing microspores at the tetrad, uninucleate or binucleate stage were cultured on various combinations of media and growth regulators.
The callus induction of D. chinensis were high when cultured on MS medium containing 1 mg L-1 2,4-D + 0.1 mg L-1 NAA and shoot regeneration of D. chinensis were high when cultured on B5 containing 1 mg L-1 TDZ + 0.1 mg L-1 NAA. Anther culture of D. barbatus in MS or B5 medium containing 1 mg L-1 TDZ and 0.1 mg L-1 NAA produced highest percentage of calli.
Nevertheless only calli from anther cultured on MS medium containing 1 mg L-1 TDZ and 0.1 mg L-1 NAA could produce shoots.
The ploidy levels of most regenerated shoots, determined by flow-cytometry, were diploid.
