CELLULAR TOTIPOTENCY IN CHRYSANTHEMUM TISSUES CULTURED IN VITRO

Abstract:
The capacity for formation of callus, root and shoot from three varieties of Chrysanthemum morifolium as a function of their origin and physiological state of the starting explant has been examined.

Excised stem, root and pedicel segments, leaf discs and apical meristem explants were grown on a defined synthetic medium. Auxins (2,4-D, NAA, and IAA), cytokinins (BA, kinetin and zeatin) and gibberellic acid GA3) were tested for their ability to induce callus and organ formation. The morphogenetic responses of the different explants varied under the same hormonal treatment. In no case there was direct regeneration of shoot buds from the mature tissue explants (stem, leaf, root and pedicel) while formation of multiple shoots were observed from apical meristem explants with cytokinin alone or along with NAA. In all the explants there were rapid development of friable calluses with auxins. Root initiation was very common, particularly with IAA. No significant morphological effects on roots was observed with the three auxins tested.

Cell suspension cultures also were established readily and maintained on the culture media found favourable for callus and bud growth. In liquid media green callus from single cells were formed which first developed roots and then shoots.
