INFLUENCE OF SEVERAL GROWTH REGULATORS AND AMINO ACIDS ON IN VITRO ORGANOGENESIS OF TORENIA AND DAUCUS

Abstract:
The effects of several plant growth regulators and various amino acids on in vitro organogenesis of Torenia fournieri Lind. and Daucus carota L. were investigated. In stem segments of Torenia, the treatment with 2,4-D resulted in nodular callus formation, while NAA stimulated root formation. The degree of bud formation was increased by an addition of cytokinins. Numerous roots were induced by treatment with NAA plus Zeatin. Effects of various amino acids were also tested on a defined medium in which nitrate (20 mM) was the only nitrogen source. The following results were obtained with stem segments of Torenia at a concentration of 5mM for all amino acids tested. L-Asparagine and L-Glutamic acid were very stimulative for both callus and bud formation. L-Glutamine, L-Aspartic acid and L-Proline had no effect. L-Serine slightly inhibited bud formation. L-Phenyl-Alanine inhibited callus formation but not bud formation. L-Lysine, L-Leucine, L-Arginine, L-Valine and L-Histidine completely inhibited both callus and bud formation. On the other hand, in a suspension culture of carrot cells, when L-Asparagine (0, 1-1mM) was added to the medium without 2,4-D, it was highly stimulative for the formation and development of embryoids. L-Glutamine, L-Arginine and L-Proline were also moderately effective at the same concentration. L-Leucine and L-Valine were inhibitory for embryoid formation.
