CLONING OF ACC OXIDASE GENE OF CARNATION AND CONSTRUCTION OF ITS PLANT ANTISENSE EXPRESSION VECTORS

Abstract:
A pair of specific primers was designed according to the cDNA sequence of DC-ACO (Dianthus caryophyllus L. ACC Oxidase) and a fragment of the gene was amplified from the genomic DNA of a carnation variety ‘Master’ by PCR. Analysis of the cloned sequence showed that it was the same as the reported sequences (AB042320.1). An antisense expression vector, named pCAM-ACO2, of the DC-ACO gene was constructed, with the antisense sequence controlled by the CaMV 35S promoter. This work is necessary for transgenic research aimed at prolonging the flowering period of carnation via antisense technology.
