APPLICATION OF PCR-BASED MARKERS IN FRAGRANT AZALEA BREEDING

Abstract:
For the purpose of fragrant evergreen azalea breeding, we obtained intersubgeneric hybrids in reciprocal cross combinations between evergreen azaleas (Rhododendron nakaharae and its hybrids) and fragrant deciduous azaleas (R. arborescens and R. viscosum). In these hybrids nuclear and organelle DNA were investigated using PCR-RFLP markers. The hybrid seedlings have nuclear ribosomal DNA (nrDNA) inherited biparentally, mitochondrial DNA (mtDNA) from seed parent and chloroplast DNA (cpDNA) from deciduous azalea regardless of cross combination. These results suggested that the chloroplast genome from deciduous azalea and the nuclear genome from evergreen azalea have compatibility in viable hybrid progenies. The application of PCR-based markers is a powerful tool to resolve complex nuclear and organelle genome hybridization mechanisms of wide cross combinations in azalea.
