FERTILITY RESTORING OF INTERSPECIFIC HYBRIDS BETWEEN LILIUM NOBILISSIMUM AND L. REGALE BY CHROMOSOME DOUBLING

Abstract:
In vitro scales of interspecific hybrids between Lilium nobilissimum and L. regale were treated with 0.5 mM (about 0.02%) colchicine to overcome pollen sterility by chromosome doubling. With an increase in the duration of colchicine treatment, the bulblet regeneration from scales decreased, whereas the tetraploid bulblets increased. In colchicine treatments for 1, 3 and 5 days, percentages of scales regenerated bulblet were 78%, 30% and 5%, while frequency of tetraploid bulblets were 2%, 15% and 33%, respectively. Pollen fertility of interspecific hybrids was 46% in tetraploid plants, but less than 1% in diploid ones. Tetraploid interspecific hybrids formed seeds containing embryo and endosperm after self-pollination. All of investigated self- progenies of the hybrids were tetraploid. These results indicate that the treatments of 0.5 mM colchicine for 3-5 days are effective in doubling chromosome for in vitro scales, and the sterility of interspecific lily hybrids can be restored by chromosome doubling.
