Abstract:
The pollen viability and longevity of chrysanthemum (Dendranthema ×grandiflorum) with small inflorescences were investigated by in vitro pollen germination.
The pollen viability of three chrysanthemum cultivars, i.e., 'rm1-2', 'rm1-3' and 'rm7-3' were 34.6%, 24.9% and 27.9%, respectively.
Pollen longevity generally lasted for about 16 days under room temperature and 35-45% relative humidity, once pollen viability under 5% was considered to be no vigor.
Pistil receptivity, either pollinated in vitro or in vivo, was detected using fluorescence microscopy.
It was found that receptivity period of the stigma maintained for 15 days since the ray florets became to colorate, and pistil receptivity reached the peak on the 11th day when the style elongated distinctly and the stigma was kicking pleat and presenting a definite angle.
Pistil development in ray florets that were emasculated only was similar to that in intact florets.
However, the development delayed 1-2 days while emasculation coupled with wiping off the petals of ray florets was conducted.
