Abstract:
Low multiplication rate is one of the problems in micropropagation of tree peony.
In the present study for shoot culture of two cultivars 'Wu Long Peng Sheng' and 'Ying Ri Hong', some factors which may influence multiplication were examined in order to set up optimal media for the multiplication of each cultivar.
Basal media modified on MS with different Ca (calcium) sources, concentration of NH4+-N (ammonium nitrate–nitrogen), NO3--N (nitrate-nitrogen) and ratio of NH4+/NO3- were tested.
Influence of sucrose (20, 30 and 40 g•L-1), 6-Benzylaminopurine (BAP 0.2, 0.5 and 1.0 mg·L-1) and Gibberellin A3 (GA3 0.2, 0.5 and 1.0 mg·L-1) on multiplication was then studied by using orthogonal design in the selected optimal basal medium.
Results showed that the decrease of the ratio of NH4+/NO3- was positive on both cultivars, not only improving multiplication rate, but releasing more plantlets from vitrificated and lightening browning degree of plantlets and culture media.
The variance analysis and visual analysis of data showed that BAP and sucrose significantly affected the multiplication.
The sequence was BAP>sucrose>GA3. The optimal medium for the multiplication of 'Wu Long Peng Sheng' was modified MS + 40 g·L-1 sucrose+1.0 mg·L-1 BAP + 0.2 mg·L-1 GA3 and that of 'Yi Ri Hong' was modified MS + 40 g·L-1 sucrose + 1.0 mg•L-1 BAP + 0.5 mg·L-1 GA3.
