PLANT REGENERATION FROM DIFFERENT EXPLANTS OF LILIUM 'TIBER' AND 'REIZAH 1'

Abstract:
The regeneration capacity of explants leaves, stem nodes, bulblet scales, filaments and pedicels of Oriental hybrids 'Tiber' or Longiflorum hybrids 'Reizah 1' were investigated to establish a genetic transformation system. The results showed that leaves were the preferred explant to regenerate plantlets by organogenesis. The filament and bulblet scale were preferred to regenerate plantlets directly. Shoot differentiation benefited from higher concentrations of BA and roots benefited from 2,4-D. The bulblets were produced in vitro directly from commercial bulb scale. Using 100 mg/L carbenicillin in the initial culture of commercial bulb scales showed good effects of sterilization. The regeneration capacity of filaments was highest when the bud was pea green and 8-10 cm in length.
