MICROPROPAGATION OF CHRYSANTHEMUM CULTIVARS - EFFECT OF COLD STORAGE ON PLANT REGENERATION IN VITRO

Abstract:
In vitro plant regeneration of chrysanthemum: cultivars ‘Reagan Sunny’ (RS) and ‘White Spider’ (WS) were investigated. Induction of morphogenesis in stem segment culture was achieved on MS solid medium supplemented with NAA and BAP (0.5 and 1.0 mgL-1, respectively). The best shoot multiplication index was observed for cv. RS. Multiplication index was subsequently decreased after long-term culture of both cultivars. The effect of cold storage treatment on index of shoot multiplication and rooting efficiency was studied. Cultures were stored at +4°C during 2, 4 and 6 months without subculturing. There was no statistical difference in shoot multiplication index after storage treatment for both cultivars. Multiplication index was increased compared to cultures constantly grown at temperature (23°C). Rooting of shoots was very successful (100%) on solid MS hormone free medium. Cold storage treatment did not influence on rooting efficiency of both cultivars. Potted chrysanthemum plants were acclimatized 53, 54, 96% for RS and 96, 50, 73% for WS after cold storage (2, 4, 6 months, respectively) and flowered. Some changes in color of flowers are noticed.
