Abstract:
In garlic tissue culture, cytokinin generally needs to be added in medium to increase seedling propagation coefficient.
However, in this study, appropriate cold treatment at 4°C was also found to have the same effect on garlic bulb propagation.
Using Chinese local garlic variety (‘Sanyuehuang’) as the material and LS as the medium, cold treatment at 4°C respectively for 0, 2, 4, 6, 8 and 10 weeks, we studied the propagation and bulbs development of garlic in stem-disc culture.
The results showed that cold treatment could enhance the propagation coefficient of garlic and accelerate garlic bulb development.
The best effects were found in cold treatment for 6 and 4 weeks, while the propagation coefficients were high, up to 7.34 and 3.85, respectively.
At the same time, the ratio of micro-bulbs development reached 89.6% and 54.7%, rooting number was 2.8 and 4.9 with the average weight of bulb 70~80 mg.
The germination ratios of micro-bulbs were correlative to their own weights, and the germination ratios would be 100% when bulb weights were over 50 mg.
