IDENTIFICATION OF GENETIC MARKERS LINKED TO THE CYTOPLASMIC MALE STERILITY GENES AND SEQUENCING SPECIFIC FRAGMENTS IN NON-HEADING CHINESE CABBAGE

Abstract:
RAPD technique was employed to detect the polymorphism between the OguCMS and its maintainer line. Two specific fragments, OPAX1800 and OPAF12600, were amplified from the OguCMS and further linkage analysis was made between OPAX1800, OPAF12600 and cytoplasmic types. Sequence analysis showed OPAX1800 contained 773 base pairs, among which 54.20% were A and T base and 42.69% were A and G base. There were more than one start and stop codons in the sequence. Nine polypeptides containing 9 to 74 amino acids were deduced from the DNA sequence. Its similarity to reported sequences was low, which indicated it was a new DNA sequence of non-heading Chinese cabbage. As to OPAF12600, it contained 57.22% A and T base and 53.04% A and G base. There were several start and stop codons in the sequence and three polypeptides containing 18, 24 and 131 amino acids were deduced from the DNA sequence. The blast results exhibited high similarity to a cDNA sequence of unknown protein in Arabidopsis thaliana. The deduced amino acid of the two fragments showed low identity to reported sequence. Two pairs of primers were designed and the RAPD markers were converted to SCAR markers.
