RAPD ANALYSIS FOR mtDNA OF OguCMS AND ITS MAINTAINER IN NON-HEADING CHINESE CABBAGE

Abstract:
Maternally inherited cytoplasmic male sterility (CMS) occurs in many plant species and is widely used to facilitate the production of hybrid seed because it eliminates the need for emasculation by hand. Meanwhile, it is a useful model for study of cytoplasm-nucleus interaction. A new OguCMS hybrid was created by asymmetric cell fusion in non-heading Chinese cabbage (Brassica campestris L. ssp. chinensis), which overcame chlorosis in the original CMS line at low temperature. Since then, much work has been done on its physiology, biochemistry and genetics. RAPD analysis was employed to study the mtDNA of OguCMS and its maintainer in non-heading Chinese cabbage. Results showed many differences between the OguCMS and its maintainer. One specific mtDNA fragment OPAG20700 was amplified from the male sterile line. Sequence analysis showed it contained 57.22% A and T base, 53.04% A and G base. It encoded one polypeptide with 225 amino acids. The sequence had high identity to AP3 gene in Brassica napus and Brassica oleracea, which indicated OPAG20700 had partial DNA sequence of AP3 gene. The access number of this sequence in the Genbank was AY623003.
