PROTEOME OF MALUS × DOMESTICA BORKH. CV. ANNURCA

Abstract:
The aim of this study was to conduct a proteomic analysis of the pseudocarp tissues of Malus × domestica Borkh. cv. Annurca. 'Annurca' is an old, high-quality apple cultivar with crisp and sweet fruits characterized by long shelf life. It is very popular in the south of Italy and represents about 60% of the entire apple yield in the Campania region. Total protein samples from the fruit pseudocarp of this cultivar were electrophoretically separated and protein species were identified by a combined matrix-assisted laser desorption/ionization – mass spectrometry and liquid chromatography – electrospray ionization tandem mass spectrometry approach. Among 303 distinct spots detected in samples, 44 species were identified and associated to 28 different genes. Energy metabolism was the functional class that was most represented in the map. In fact, within this class we detected the expression of enzymes of glycolysis, the pentose phosphate pathway, respiration, fermentation and ATP-proton motive force. Difficulties in the identification were related to the poorly characterized Malus genome; so, this reference map may serve as a tool in future studies aimed at the evaluation of changes in protein repertoire during interactions with pathogens.
