PRELIMINARY STUDIES OF IN VITRO CLONING OF DRACAENA (DRACAENA SANDERIANA)

Abstract:
Dracaena is an important foliage plant and is commercially propagated by vegetative means. Propagation by terminal cuttings produces a single plant. A mature dracaena plant produces 3-4 plants through cuttings. The present investigation was undertaken to develop a viable protocol for in vitro cloning of dracaena to bypass the slow rate of multiplication of traditional methods. Stem cuttings containing single nodes were used as explants and tested for surface sterilization, proliferation and root formation studies. Our results indicate that a 5 minute surface sterilization followed by initiation on medium with BA and IBA allowed initiation of 90% of explants. Further growth on BAP (2 mg/L), NAA (0.5 mg/L) and GA3 (5 mg/L) produced the best stem and leaf growth but did not allow multiplication of shoots. Preliminary rooting studies found 80% rooting on IBA (3 mg/l) + GA3 (2 mg/L). Additional studies are needed to improve the shoot multiplication of this species.
