ANTI-INFLAMMATORY COMPOUNDS FROM MEDICINAL PLANT RUTA GRAVEOLENS

Abstract:
Twelve plant extracts that are traditionally used for the treatment of rheumatism, arthritis and other inflammatory conditions in the traditional medicine of India, were evaluated for their anti-inflammatory properties. Of these the 50% methanolic extract of Adonis vernalis was found to inhibit the tumor necrosis factor-α (TNF- α) production significantly by the whole blood cultured cells. Similarly, the methanol extracts of Quassia amara, Ruta graveolens and Boswellia serrata were observed to inhibit the nitric oxide (NO) production by the murine macrophage cells challenged with 1µg/ml LPS when incubated simultaneously. On preincubation with the plant extract and then LPS challenge after washing the macrophage cells, only the Ruta and Quassia extract were found to inhibit the NO production (73% and 50%, respectively). The methanolic extract of the Ruta graveolens plant showing maximum NO inhibition on further fractionation by solvent extraction and reverse phase HPLC and two active compounds Rg001 and Rg002 were identified. These two compounds were found to be more potent inhibitors of NO and IL-1β. The compounds Rg-001 and Rg002 were respectively characterized as 3-(1,1-dimethyl-allyl)6-hydroxy-7-methoxy-chromen-2-one and 3-(1,1-dimethyl-allyl)6-hydroxy-chromen-2-one (patent pending) by ESI-MS, MALDI-TOF, FT-IR and NMR.
