EFFECT OF GENOTYPE ON CALLUS FORMATION AND ORGANOGENESIS IN VITIS

Abstract:
‘Thompson Seedless’, ‘Cardinal’ grape cultivar and ‘Fercal’, ‘Kober 5BB’ grapevine rootstock were propagated in vitro conditions via shoot tip culture. Leaf laminae, petiole and internode explants were excised from plantlets and cultured in MS medium supplemented with different BAP and 2,4-D combinations for callus formation and organogenesis. Media containing 1 mg/L BAP in combination with 0.1 mg/L or 1 mg/L 2,4-D was found to be the best for callus formation of all grape cultivars while 0.2 mg/L BAP in combination 0.1 mg/L 2,4-D of all grapevine rootstocks. Organogenesis was not observed when the medium was supplemented with 2,4-D while it was induced with BAP for all genotypes. Fercal rootstock gave the best results in callus formation and organogenesis when compared to other genotypes.
