Abstract:
The purpose of this study was to establish culture medium requirements for micropropagation of Vitis vinifera L. cv.
Listan blanco.
However, some problems such as, contamination, phenolic exudates and hyperhydricity symptoms were detected and solved.
Cultures were initiated from apical meristem collected from mature plants during the spring.
To avoid contamination rate in meristem culture of this variety, fungicide solution (1% benomyl, 15 min) should be used and also a small amount (0.3 %) of surfactant such as Tween or Teepol should be added to the Cl2Hg. Most satisfying results were obtained using a solidified and a modified WP enriched with KNO3 plus GA3, BAP and IAA. Moreover, the prevention of the browning phenomenon is essential to avoid meristem necrosis during establishment of these tissue in culture.
Whole shoots were used for in vitro shoots multiplication and best results were achieved with WP.1 medium plus BAP and IAA or NAA. Shoots rooted without symptoms of chlorosis or necrosis in half-strength WP medium plus NAA or IBA. On the other hand, addition of gerlite in solid medium led to a significant decrease in hyperhydricity during the rooting stage, without showing any significant detrimental effect on the shoots’ number of in vitro plantlets produced for this cultivar.
