MORPHOLOGY, POD ISOZYME AND RAPD ANALYSES OF PLANTS REGENERATED FROM EMS-TREATED SHOOT TIPS IN 'DAWUXING' LOQUAT

Abstract:
Shoot tips of in vitro plants of loquat (Eriobotrya japonica Lindl. ‘Dawuxing’) treated with ethylmethane sulfonate (EMS) to induce mutation and morphological, biochemical and molecular changes, were identified. EMS treatment caused changes in shoot height, stem node length and leaf shape. The overall frequency of morphologic variation was about 18.5%, ranging 8.3–33.3%, the greatest being 30% after treatment with 0.9% EMS for 0.5–1.5h. Changes in POD isozyme spectrum were found in the plants with morphologic variants. An extra POD isozyme band, whose Rf value was 0.2959, was identified after treatment with 0.1% EMS for 0.5h, 0.3% EMS for 0.5h or 0.3% EMS for 1.0h. Treatment with 0.9% EMS for 1.5h caused changes in POD isozymes and the Rf 0.0723 band disappeared. The overall frequency of POD isozyme spectrum variation was about 7%, the highest POD isozyme spectrum variation rate reaching 17% in the treatment with 0.9% EMS for 1.5h. Changes were also detected in RAPD amplification profiles in the plants from EMS treatment. There were 8 (4.6%), 12 (6.8%) and 13 (7.0%) plants with variant RAPD amplification profiles.
