MICROPROPAGATION IN FOUR POTATO GENOTYPES AND SELECTION ON VITROPLANTS SIZE AS A SURVIVAL EX VITRO ESTABLISHMENT

Abstract:
In the potato micropropagation, the addition of 116 mM sucrose to the cultivation medium increases the Fresh Weight (FW) and Dry Weight (DW) of in vitro generated plants; this response favors the hardening during the acclimatization. In the greenhouse, the hardening is reached in 28 days; the change from heterotrophic to autotrophic condition is best in medium plants (31 to 50 mm). The best survival rate was obtained in the Northern variety (100%) and 91–10–1 (93%) clone. There were no statistical differences in clones 91–18–2 1 and 91–32–5 1 (79%). The nutrimental state in vitro and ex vitro is decisive for the material establishment in greenhouse; the foliar NPK and Ca concentrations were inside of established levels, but the microelements concentrations like Mg and Cu were not enough. In clones 91–10–1, 91–32–5 and in the Northern variety, the Mn, Cu Fe and Zn microelements foliar analyses were classified as appropriate.
