DIFFERENCES IN MICROPROPAGATION OF VEGETATIVE ROOTSTOCK (GF677) AND OTHER ALMOND SEED GENOTYPES

Abstract:
In this research, a suitable media for micro propagation of GF677 rootstock and almond seed genotypes was determined. The results showed that the most suitable medium for GF677 was different for seed genotypes in different stages. In the proliferation stage of GF677, modified MS medium supplemented with 0.7 mg/L BA and 0.01mg/L NAA showed the best result but the proliferation of seed genotypes was obtained in the same MS medium with 1mg/L BA And 0.01 mg/L NAA. The optimum elongation occurred for GF677 in modified KNOP medium supplemented with 0.1mg/L BA and for seed genotypes with the same BA concentration but in modified MS medium. GF677 rooted successfully (up to 80%) in LS medium supplemented with 0.3mg/L NAA in darkness. But rooting of seed genotypes had some special problems. It can be concluded that as stem cuttings of seed genotypes cannot be rooted easily by conventional propagation methods, it is difficult to root them in micropropagation as well.
