EFFECTS OF TEMPERATURE ON THE GROWTH AND DORMANCY OF TISSUE-CULTURED HERBACEOUS PEONY SHOOTS

Abstract:
The influence of temperature (15°C, 20°C, 25°C) on the growth and development of non-cooled (dormant) and cooled (non-dormant) herbaceous peony ‘Jadwiga’ shoots grown in vitro was investigated. In the herbaceous peony shoots that were propagated in the high temperature (20°C, 25°C) the induction of dormancy was observed. The rate of growth and the rate of multiplication were reduced and senescence and death of the leaves took place. For the experiment, one set of peony shoots was cooled at +4°C (2 months) and the second part of shoots was grown continually at 25°C. The cooling treatment reactivated further growth and development of peony shoots at all temperatures. The cooled explants grown at 15°C, produced the highest numbers of shoots and leaves during the first subculture after cooling. The fresh weight of shoots was also very high for this subculture. The non-cooled explants produced lower numbers of shoots and leaves compared with the cooled shoots. Shoot formation and emergence of leaves decreased with the increase of temperature for cooled and non-cooled explants. The multiplication rate of cooled and non-cooled explants decreased in the third subculture independently of temperature.
