IDENTIFICATION OF TOMBUSVIRUSES ISOLATED FROM LIMONIUM SINUATUM ON THE BASIS OF HOST RANGE, SEROLOGICAL AND MOLECULAR STUDIES

Abstract:
Since 1995 the Dutch Plant Protection Service occasionally received samples of Limonium sinuatum for diagnosis. Submitted plants showed growth reduction and distortion. After mechanical inoculation, herbaceous test plants showed virus-like symptoms. In electron microscopy isometric particles with a diameter of about 30-32nm resembling those of tombusviruses, were observed. For further identification host range, serological and molecular studies were performed. Mechanical inoculation to 15 different plant species did not reveal significant differences between the isolates. However, by immuno-electron microscopy using antisera to 14 different tombusviruses or tombuslike-viruses, four groups could be distinguished. Subsequent sequence analysis of RT-PCR products covering the coat protein gene, provided additional data which indicated that six of the isolates belong to one of the following tombusvirus species: Petunia asteroid mosaic virus, Grapevine Algerian latent virus or Cymbidium ringspot virus. The seventh isolate represents a new virus species, for which the name Limonium flower distortion virus is proposed.
