Abstract:
We are constructing a physical framework for peach, which is a model fruit tree for the Rosaceae species.
Currently the physical framework is composed of 1,128 contigs containing 8,508 BAC clones.
Conservative estimates indicated these contigs cover at least 53% of the peach genome.
Based on core genetic marker hybridizations, 168 contigs were anchored onto an integrated physical/genetic map.
The 47 positioned contigs were localized on linkage group 1, which represents the most marker-saturated chromosome of peach.
To verify the physical framework, we used 673 ESTs of the peach unigene set (PP_LE) and 64 cDNA (LF) clones, which were incorporated into the framework by random fingerprinting.
At this juncture, more than 12% of BAC clones in the peach FPC database have hybridization hits (i.e. core genetic markers, ESTs, cDNAs). The physical framework for peach is located at the Genome Database for Rosaceae at www.genome.clemson.edu/gdr/.
