ORNAMENTAL PEACH AND ITS GENETIC RELATIONSHIPS REVEALED BY INTER-SIMPLE SEQUENCE REPEAT (ISSR) FINGERPRINTS

Abstract:
Ornamental peach (Prunus persica (L.) Batsch) has many outstanding and popular cultivars. Although many of them have been widely used in urban landscape and gardens, the genetic relationship among these ornamental peach cultivars is not clear, which limits the further studies of its molecular systematics. ISSR markers were applied to analyze genetic relationships among 16 ornamental peach taxa. All 100 available primers were screened using two cultivars and ten primers with the most polymorphic bands were selected for this study. A total of 132 useful markers between 300 to 1,400 base pairs were generated. Among them, 62% of bands were polymorphic markers. The average number of markers for each taxon was 80. Prunus davidiana (Carr.) was apparently an outgroup to P. persica and its two hybrid cultivars were grouped to a cluster, with 69% bootstrap support, which indicated the pedigree of P. davidiana involved in ornamental peach cultivars and that these cultivars do have apparent genetic distance to P. persica cultivars. The average genetic distance between five upright growth habit cultivars was 0.091 and the average genetic distance between three cultivars with dwarf growth habit was 0.101. These results demonstrated that ISSR markers are a useful technique to reveal group (different pedigree and different growth habit) genetic relationships of ornamental peach. The proper protocol and ISSR-PCR system for ornamental peach established in this study could help future ornamental peach germplasm discrimination, conservation, and new cultivar development.
