Abstract:
We developed 24 PCR-RFLP markers for strawberry (Fragaria × ananassa Duch.) cultivar identification.
We used these co-dominant markers to successfully distinguish 65 commercial cultivars.
The markers were also capable of distinguishing closely related strains (96 progeny of a selfed ‘Sachinoka’ line). The results were highly reproducible across DNA extraction methods, organs, and researchers.
The stability and simplicity of the PCR-RFLP analysis makes it a valuable tool for identifying strawberry cultivars.
In addition, we investigated the inheritance of these markers using selfed lines of ‘Sachinoka’, T’ochihime’, ‘Nyoho’, and ‘Cesena’, and checked whether it was consistent with Mendelian law.
